چکیده

Cancer is one of the most common diseases in world. Th e transcription factor NF-κB plays a key role various physiological processes including immune response, cell proliferation, apoptosis and inflammation. Due to participation NF-kappa B signaling pathways carcinogenesis, angiogenesis, tumor resistance chemo- radiotherapy, considered be ideal target for pharmacological treatment cancer. paper presents literature review RSCI, PubMed, Scopus, Web Science, Chemical Abstracts NCCN databases other open access data. The carcinogenesis inhibitors were selected from all found. Inhibitors with antitumor activity analytical interest include Bruton tyrosine kinase (BTK) inhibitors, cellular proteins (c-IAP), proteasome inhibitor translocation nucleus. authors analyzed products diff erent development stages recorded varying degrees. are promising drug candidates, but since involved biological broad spectrum action can cause side eff ects, selective these compounds explored.

NF-κB Inhibitors from Eurycoma longifolia

The roots of Eurycoma longifolia have been used in many countries of Southeast Asia to alleviate various diseases including malaria, dysentery, sexual insufficiency, and rheumatism. Although numerous studies have reported the pharmacological properties of E. longifolia, the mode of action of the anti-inflammatory activity has not been elucidated. Bioguided isolation of NF-κB inhibitors using an...

Inhibition of Canonical NF-κB Signaling by a Small Molecule Targeting NEMO-Ubiquitin Interaction

The IκB kinase (IKK) complex acts as the gatekeeper of canonical NF-κB signaling, thereby regulating immunity, inflammation and cancer. It consists of the catalytic subunits IKKα and IKKβ and the regulatory subunit NEMO/IKKγ. Here, we show that the ubiquitin binding domain (UBAN) in NEMO is essential for IKK/NF-κB activation in response to TNFα, but not IL-1β stimulation. By screening a natural...

Small molecule inhibitors of ceramidases.

The equilibrium between the pro-apoptotic ceramide and pro-vital sphingosine-1-phosphate is considered to be decisive for cell death or survival. The different ceramidases thus play key roles in cell fate and might offer attractive targets for pharmacological intervention. Although until recently only moderately active inhibitors have been described, first in vivo experiments suggest activity a...

Small Molecule Inhibitors of CXCR4

CXCR4 is a G-protein-coupled receptor involved in a number of physiological processes in the hematopoietic and immune systems. The SDF-1/CXCR4 axis is significantly associated with several diseases, such as HIV, cancer, WHIM syndrome, rheumatoid arthritis, pulmonary fibrosis and lupus. For example, CXCR4 is one of the major co-receptors for HIV entry into target cells, while in cancer it plays ...

Allosteric small-molecule kinase inhibitors.

Small-molecule kinase inhibitors are invaluable targeted therapeutics for the treatment of various human diseases, especially cancers. While the majority of approved and developed preclinical small-molecule inhibitors are characterized as type I or type II inhibitors that target the ATP-binding pocket of kinases, the remarkable sequential and structural similarity among ATP pockets renders the ...
